Investigation of Apoptotic Effect of Klotho Protein on Human Colorectal Cancer Cells via TRAIL Death Receptors

Klotho is a transmembrane protein which deficiency has pleiotropic effects in number of aging-related disease processes and various cancers. In some recent studies about klotho disorders, it been shown to effective cancers including lung, liver, breast, kidney, colon. TNF-related apoptosis-inducing ligand (TRAIL), TNF family molecule, cytokine that stimulates apoptosis through death receptors many cancer types therefore attracts attention for tumor therapies preclinical models. Interaction with TRAIL create an apoptotic effect treatments by reducing the proliferation cells. this study, was aimed investigate exogen administration on cell viability (TRAIL1 TRAILR2) human healthy colon cells (CCD 841 CoN) TRAIL-resistant colorectal (caco2). For purpose, were treated different concentrations 24 48 hours. To determine viability, proliferation, evaluations WST-8 Real-Time qRT-PCR analysis performed. Our results showed increase concentration did not have significant cells, whereas decreased apoptosis-resistant Relative gene expression TRAIL1 TRAILR2 increased applied line caco-2. Therefore, targeting ligand-receptors induce can be considered as potential therapeutic approach therapy.
